1. Aim of classes
This practical focuses on getting familiar with the course of basic metabolic processes accompanying the neutralization of pollutants in soil and water environment.
2. Procedures
1. Hydrolysis of starch
1. Dividing a starch-containing agar plate into four section .
2. Labeling each with the bacterium to be inoculated .
3. Using an aseptic technique, streak the respective bacteria onto the plate within the section.
4. Incubating the plate for 24 to 48 h at 37°C
5. Placing several drops of Lugol's solution on each of the line streaks on the starch agar plate and after few minutes inspecting it for the color development if blue color appears the starch is still present in medium(it is not hydrolyzed); if a colorless or yellow brown area is seen, starch is hydrolyzed.
2. Hydrolysis of fats
1. Dividing a fat-containing agar plate into four section
2. Labeing each with the bacterium to be inoculated
3. Using an aseptic technique, streak the respective bacteria onto the plate within the section
4. Incubating the plate for 24 to 48 h at 37°C
5. Placing several drops of CuSO4 solution on each of the line streaks on the starch agar plate and then inspecting it for the color development if agar plate contains lipase-producing microorganisms, a green area surrounding the growth zone can be seen (copper ions form green-colored complexes with free fatty acids).
3. Hydrolysis of proteins
1. Dividing a gelatin-containing agar plate into four section
2. Labeling each with the bacterium to be inoculated
3. Using an aseptic technique, streak the respective bacteria onto the plate within the section
4. Incubating the plate for 24 to 48 h at 37°C
5. Inspecting agar plate - if the area surrounding the colony appear clear, the gelatin molecules are digested.
4. Hydrolysis of lactose
1. Dividing a agar plate containing lactose and bromocresol purple into four section
2. Labeling each with the bacterium to be inoculated
3. Using an aseptic technique, streak the respective bacteria onto the plate within the section
4. Incubating the plate for 24 to 48 h at 37°C
5. Inspecting agar plate - if the area surrounding the colony appear yellow, the lactose molecules are digested
